A C18 (octadecylsilane) column is packed with porous silica particles whose surface carries 18-carbon chains. The chains form a hydrophobic surface that holds peptides according to their hydrophobicity.
Pore size matters for peptides: columns with pores of about 100–120 Å suit small peptides, while wide-pore columns of about 300 Å give larger peptides and small proteins access to the bonded surface. C8 and C4 phases, with shorter chains, are used for very hydrophobic or larger molecules.
